A total of 4,938,717 patients with schizophrenia or major depressive disorder were studied, and 2,091,766 patients were randomized to receive Seroquel SR (Qelzefin extended release) or a placebo. Patients were assessed at baseline and 2, 6 and 12 months, at the end of the study and at the end of the follow-up period. Treatment outcomes (seroquel withdrawal syndrome, number needed to treat, response rate, percentage of patients achieving a defined therapeutic dose) were evaluated at each follow-up visit. The main endpoint of the study was the frequency of withdrawal. After the primary endpoint was reached, the rate of treatment withdrawal was reduced by 22.3% (95% confidence interval [CI], 20.1–24.9%), and the rate of treatment withdrawal decreased by 19.8% (95% CI, 13.5–24.0%) in patients randomized to the Seroquel SR compared with those in the placebo (P<0.001). The rate of treatment withdrawal was also reduced by 19.8% (95% CI, 13.5–24.0%) in patients randomized to the Qelzefin extended release compared with patients in the placebo (P<0.001). Seroquel withdrawal syndrome was also reduced by 23.4% (95% CI, 23.6–24.0%) in the Seroquel SR compared with those in the placebo (P=0.008). There was no significant difference in the rate of treatment withdrawal between the Seroquel SR and placebo (P=0.073). The rate of treatment withdrawal was reduced by 15.9% (95% CI, 10.0–17.6%) in the Seroquel SR compared with those in the placebo (P<0.001). The rate of treatment withdrawal was decreased by 19.8% (95% CI, 13.5–24.0%) in the Seroquel SR compared with those in the placebo (P<0.001). There was no significant difference in the rate of treatment withdrawal between the Seroquel SR and placebo (P=0.5) and between the Seroquel SR and Qelzefin extended release (P=0.9).

The Qelzefin extended release dose was 1.25 mg/kg/day in the Seroquel SR group and 1.25 mg/kg/day in the placebo group (P=0.006). The rate of discontinuation due to adverse events in the 2, 9, and 12-month periods were similar between the two groups (P<0.001).
